No you do not need to manually click the primary slot you placed on your hot bar. Once the key is bound to it, hitting that key will actually activate the effect on the weapon
So say stopsong is bound to ~ and say the primary slot is bound to %..
Pressing ~, %, will actually stopsong and cast nivs melody of preservation
